Gerbera (Gerbera jamesonii) belongs to the Asteraceae. It is one of the most popular ornamental flowers worldwide and used both as cut flower and potted plant. They have some characters such as color, floral diameter, stem length, and vigor, which make this plant of commercial importance. Therefore, Gerbera tissue culture is essential to provide seedling for production. The purpose of this study was to observe the influence of plant growth regulators on in vitro propagation of Gerbera. The results showed that: i) all the samples had formed callus after 21 days of culture and the best callus formation obtained when the thin cell layer of Gerbera was treated with 0,5-0,5, and 0,5-1 mg/l of NAA – BAP
ii) the best medium for shoot formation from callus was MS1 (8,5 shoots per explant), the best medium for shoot formation from callus was MS1 (8,5 shoots per explant), thus reducing costs and inhibitory effect of high concentrations of macronutrients
iii) MS1 medium supplemented with 1 mg/l kinetin was the best for shoot multiplication (5 shoots per explant)
iv) MS1 medium supplemented with 1 mg/l NAA was the best for root and plantlet formation.Hoa Đồng tiền có tên khoa học là Gerbera jamesonii, thuộc họ Cúc
là loại hoa phổ biến trên thế giới, sử dụng cho cả trồng chậu và cắt cành
có giá trị thương mại cao bởi các đặc tính như màu sắc, đường kính hoa và sức sống. Vì vậy, nuôi cấy mô là rất cần thiết để cung cấp đủ nhu cầu giống cho sản xuất. Mục đích nghiên cứu này là khảo sát ảnh hưởng của các chất điều hòa sinh trưởng đến quá trình nhân giống in vitro cây hoa đồng tiền. Kết quả cho thấy nồng độ kết hợp giữa NAA - BAP ở 2 mức độ 0,5-0,5 và 0,5-1 mg/l tối ưu nhất cho quá trình hình thành mô sẹo từ mẫu lớp mỏng cuống lá hoa Đồng tiền in vitro. Nồng độ môi trường ở MS1 bổ sung kinetin 0,5 mg/l đảm bảo sự phát triển bình thường của chồi (8,5 chồi/mẫu) từ mô sẹo lát mỏng tế bào hoa Đồng tiền, giúp giảm chi phí nuôi cấy và giảm sự ảnh hưởng ức chế của đa lượng ở nồng độ cao trong môi trường đến mẫu chồi in vitro. Môi trường MS1 có bổ sung kinetin nồng độ 1 mg/l cho chỉ số nhân chồi cao nhất (5 chồi/mẫu). Môi trường MS1 có bổ sung NAA nồng độ 1 mg/l cho khả năng hình thành rễ và cây hoàn chỉnh tốt nhất.
