In black tea processing industry, polyphenol oxidase is added in fermentation production process to improve the quality of tea. In this study, the authors conducted the cloning and sequencing gene encoding the polyphenol oxidase (PPO gene) from three species: Trung Du, LDPI and Keo Am Tich. The length of PPO gene is a appropriatly 1.8 kb, encoding 599 amino acids. The results of PPO gene analysis showed that Trung Du species had 13 nucleotide variants as compare with other species and China species. Keo AM and LDPI had 4, 5 nucleotide variants as compare with other varieties, respectively. The variations of nucleotide lead to change amino acids of the important function domains of the PPO. At the signal peptide sequence, Keo Am Tich species has a amino acid change at position 30 (1-+ T) and Trung Du species has four amino acid changes at positions 26 (K-+Q), 40 (S-+P), 42 (N-+K), 60 (L-+Q) as compare with other species. Analysis of amino acid sequence in the domains associated with the functional areas are relatively tea conservative, domain 2 (from amino acid 336 to 378) is the most conservative, domain 1 (from amino acid 183 to 240) and domain 3 (from amino acid 531 to 567) has two amino acid changes at positions 212 (F-+H), 557 (R-+T) in only Trung Du species. The variants may affect the activity of the PPO, therefore the authors need more studies of PPO.
