In black tea processing industry, polyphenol oxidase is added in fermentation production process to improve the quality of tea. In this study, the authors conducted the cloning and sequencing gene encoding the polyphenol oxidase (PPO gene) from tea variety TRI777. PPO gene obtains 1800 bp in length, encoding 599 amino acids. The comparison the nucleotide and amino acid of PPO gene from tea variety TRI777 and remaining sequences in Genbank (EU787433, EF650016, EF650017, EF623826, EF635860, JX465712, GQ129142, FJ210643 va DQ513313) showed 98,2-100 percent degree of identity in nucleotides, 97,1-100 percent in amino acid, respectively. The nucleotide sequence of PPO have fours different nucleotide positions (541 G-A, 797 T-C, 1122 C-T, 1359 A-G) compare with the remaining sequences. The nucleotide differences were found to result in 2 amino acid substitutions (181 D - N, 266 V - A). Compare result showed deductive .amino acid sequence in tea variety TRI777 are high conservative, not found different in important Cu binding domain of PPO: domain 1 (183-240), domain 2 (336-378) and domain 3 (531-567). The isolated PPO gene is a good starting material for further elucidating the function of PPO.
