Objective: 1) To compare quality of specimens between two cytological testing methods Liqui Prep and classical Pap smear. 2) To assess diagnostic accuracy by comparing the results obtained by two methods Liqui Prep and classical Pap smear with histopathological diagnostic result. Subjects and methods: This is a cross-sectional descriptive comparative study conducted on 60 women who have received gynecological check-up at the National ONGYN Hospital and tested by two methods Liqui Prep and classical Pap smear from November 2012 to January 2013. Results: The rates of well-prepared specimens of Liqui Prep and classical Pap smear were 79.9 percent and 63.3 percent, respectively. The rates of specimens felt below standard of classical Pap smear was significantly (p0.05) higher than Liqui Prep (15.0 percent to 6.7 percent, respectively). By classical Pap smear, false negative rate among the LSIL cases was 11.0 percent, while false positive rate was 17.3 percent. The false positive rate among the HSIL cases was 16.7 percent. The the Liqui-Prep, false negative rate among the LSIL cases was 8.0 percent while false positive rate was 9.0 percent. The false positive rate among the HSIL cases was 10.0 percent. Conclusion: Liqui-Prep method provides higher quality of specimens with higher sensitivity compared to the classical Pap smear.
