Maize (Zea mays L.) is an important food crop and cultivates widely in the world. Study of plant regeneration system nom immature embryos, that has high regeneration nequency, is an important prerequisite for genetic engineering of maize. In this study, the research is carried out on 16 Vietnam maize lines. Suitable sizes of immature embryos used as material sources for production of calli are nom 0.7 to 1.5 mm. The N6 medium supplemented with 2,4-D (lor 2 mg/l)
AgN03 (10 or 15 mg/l)
100 mg/l casein hydrolysate
1.15 g/l L-proline
20 g/l sucrose
and 10 g/l glucose gave the best callus production ability. The best medium for shoot regeneration is N6 basic medium supplemented with 60 g/l of sucrose and nom 0.2 mg/l BAP with regeneration nequencies nom 52 to 81.67 percent. The 1/2 N6 medium supplemented with NAA (0.5 mg/l) gave highest root formation ratios (41.1246.67 percent). H240, H264, P95, 2019, 2021 have the high regeneration ability and can be the good materials for gene transfer research in Vietnam maize.
