Inactivated proteins caused many human diseases. Protein therapy is one of the therapies for many diseases related to a defected protein or inhibitor of a biological process to stop the growth of the tumor. Here, the authors present the results of improved recombinant cell-penetrating peptides (rCPPs). These rCPPs can serve as new tools for delivering protein in biology and medicine applications. the authors cloned the CPP in-frame with' green fluorescent protein (rCPP - GFP). the authors were able to express and purify full-length rCPP - GFP fusion proteins. The results showed that, each purified r.protein was tested in Hela and Jurkat cell lines for their transduction efficiency and translocation. The purified rCPPs - GFP fusion proteins have biological activity as good as HIV virus derived TAT - EGFP peptides. The expressed rCPPs-EGFPs were able to translocate across both the cytoplasm and nucleus, thus, these intracellular regulatory proteins can be used as protein transporters for therapeutic purposes.
