Drought and salinity are among the most important abiotic stress factors limiting food production. In the context of climate change, drought and high-salt are challenges in agricultural production and the main reason for food insecurity. DREB/CBF (dehydration-responsive element-binding protein/C-repeat-binding factors (CBFs) transcription factors including DREB1s and DREB2s specifically interact with the, DRE/CRT cis acting element and control the expression of many stress-inducible genes in Arabidopsis. In this study, coding sequence of OsDREBlA was cloned in expression vector pGEX-4T to get a recombinant plasmid pGEX-4T- OsDREBlA. Fusion protein GST-OsDREB1A was over-expressed in E. coli, strain DE3 (Rossetta) by IPTG (1 mM) induction and maximum level of expression was observed after two hours of induction. Purified protein, GST- OsDREBlA was donned using Glutathione Sepharose 4B arccording to GST Gene Fusion System. Using band shift assay, the article determine that purified recombinant protein OsDREB1A bound specifically to DRE sequence localised on promoter of stress inducible gene, JRC2606 (Glutamate dehydrogenase-like proteins).
