The raffinose family of oligosaccharides (RFOs) accumulating in plant tissues are thought to playa role in the desiccation tolerance under osmoprotectant machanism and thereby, play an important role in in drought, high salt and cold stresses tolerance in plants. Gene encoding galactinol synthase (GOLS), which catalyses the first step in the biosynthesis of RFOs from UDP - galactose, were identified and cloned from many plants such as tomato Lycopersicon escu/entum Mill. cv Moneymaker (LeGOLS-1), Thellungiella halophilư (ThGOLS), Arabidopsis thaliana (AtGOLS), Coffea Arabica L.(CaGOLS)... Overexpression of AtGOLS2 in transgenic Arabidopsis caused an increase in endogenous galactinol and raffinose, and showed reduced transpiration from leaves to improve drought tolerance. In this study GOLS gene were iselated san sequencing reduced from Moc Tuyen rice drought cDNA library. As a result, ORF (Open Reading Frame) of Moc Tuyen OsGOLS consist of 1026 bp coding 342 amino acid. Analysis of its deduced amino acid sequence indicated that this protein contains the serine phosphorylation site at position 260 and the ASAAP pentapeptide hydrophobic domain characteristic at C-Terminal of all known GOLS protein. Sequence alignment of Moc Tuyen OsGOLS and homolog GOLSs from different species shown that Moc Tuyen OsGOLS had striking homology with Japonica OsGOLS1, ZmGOLS1 and AtGOLS3.
