Chymopapain (EC. 3.4.22.6) is a cysteine endopeptidase, it was isolated from the latex of the unripe fruit of the papaya (Carica papaya L., Caricaceae ). The authors extracted and refined to higher purificatory level of chymopapain (2.31 U/mg) for the pharmaceutical preparations. A process which permits the crude scale isolation of large amounls of homogeneous populations of Chymopapain, free from cross-contamination and other proteins, from either papaya latex or crude chymopapain preparations. This process comprises mixing the papaya latex or crude chymopapain with some buffers and the use of the precipation technics as the isolated and purified wIth NaCl and (NH4)2SO4 and gel filtration technics, the proteolytic activities of refined chymopapain is 1,717.80 nk/mg protein that was increased 3.2 fold. Chymopapain lyophilized powder for injection was created with the excipient mannitol, dextran in the flour mixture, and in the process of conducting experiments. Chymopapain-containing compositions are also disclosed, as is a standardized, enzyme assay for a pharmaceutical preparation of chymopapain could be used in chemonucleolysis for the treatment of sciatica. The results suggested that proteolytic activity of the initiation extract is 536.24 nk/mg protein. The specific activity of experimental pharmaceutical chymopapain (1.65-1.5U) was compared with level of standard preparation (1.75-1.5U).
